醫(yī)學論文范文:6-OHDA誘導大鼠不同腦區(qū)星形膠質(zhì)細胞反應(yīng)差異的研究
【摘要】 目的:觀察大鼠不同腦區(qū)中的星形膠質(zhì)細胞對6-羥多巴(6-OHDA)局部注射反應(yīng)的差異。方法:將6-OHDA局部注入大鼠腦內(nèi)黑質(zhì)致密部(SNc)、紋狀體(CPu)、腹側(cè)被蓋區(qū)(VTA)和額葉聯(lián)合皮層(FrA),用免疫組織化學方法觀察這些腦區(qū)中星形膠質(zhì)細胞的變化。結(jié)果:6-OHDA注入1 d后各腦區(qū)星形膠質(zhì)細胞反應(yīng)較輕,7 d后反應(yīng)明顯,并持續(xù)至14 d;各組相比較以SNc組和VTA組反應(yīng)明顯,而CPu組和FrA組反應(yīng)較輕。結(jié)論:大鼠不同腦區(qū)中的星形膠質(zhì)細胞對6-OHDA局部注射后的反應(yīng)存在差異。

[Abstract] Objective:To observe the reactions of astrocytes in different brain regions in rat model induced by 6-OHDA. Methods:The reactive astrocytes in substantia nigra compact part(SNc), caudate putamen(CPu), ventral tegmental area(VTA)and frontal association cortex(FrA) induced by 6-OHDA were detected by immunohistochemical staining. Results:The reactions of astrocytes were hardly detected on day1 after injecting 6-OHDA. From day 7 on wards the reactions were significant. We also found that the reactions of astrocytes in SNc and VTA were more remarkable than that in CPu and FrA. 動物模型常用的神經(jīng)毒劑,能選擇性損傷單胺能神經(jīng)元[1]。將其注入黑質(zhì)致密部(SNc)、紋狀體(CPu)或前腦內(nèi)側(cè)束時,會引起黑質(zhì)DA能神經(jīng)元的大量退變死亡及紋狀體內(nèi)的DA水平下降。在6-OHDA誘導的PD模型及PD患者腦中都發(fā)現(xiàn)CPu和SNc腦區(qū)出現(xiàn)明顯的膠質(zhì)細胞反應(yīng)。將6-OHDA局部注入腹側(cè)被蓋區(qū)(VTA)或額葉聯(lián)合皮層(FrA)能否同樣誘導其中DA能神經(jīng)元胞體及末梢的退變死亡,以及能否激活其內(nèi)的星形膠質(zhì)細胞?目前未見詳細的研究報道。本實驗將6-OHDA局部注入大鼠SNc、CPu、VTA和FrA腦區(qū),觀察這些腦區(qū)中星形膠質(zhì)細胞反應(yīng)的情況,借以探討不同腦區(qū)中的星形膠質(zhì)細胞對6-OHDA反應(yīng)的差異性,為進一步研究星形膠質(zhì)細胞參與DA能神經(jīng)元及其末梢選擇性損傷的機制及臨床治療PD等神經(jīng)退行性疾病提供一定的實驗依據(jù)。
1 材料與方法
1.1 實驗動物和主要試劑 成年雄性SD大鼠48只,體重220 g左右(南通大學實驗動物中心提供)。6-OHDA(Sigma公司),小鼠抗GFAP(Chemicon公司),HRP標記的山羊抗小鼠抗體(Chemicon公司),二氨基聯(lián)苯胺(DAB)(Sigma公司)。
