醫(yī)學論文范文:大鼠股骨骨不連模型不同位點骨形態(tài)發(fā)生蛋白2基因的表達
【摘要】 目的: 觀察大鼠股骨骨不連模型修復過程中骨形態(tài)發(fā)生蛋白2(bone morphogenetic protein2,BMP2)在各個時間段的表達情況,初步探討單一生長因子對骨不連修復的影響。方法: SD大鼠分別于取材前35天、28天、21天、14天、7天、3天和1天建造股骨骨不連模型,同一天處死大鼠取材,RTPCR技術檢測各個樣本的BMP2基因的表達情況。結(jié)果: 在大鼠股骨骨不連修復過程的不同時間,BMP2基因表達在中心區(qū)和外周區(qū)存在差異,中心區(qū)BMP2基因表達滯后,表達的增幅及峰值低于邊緣區(qū)。結(jié)論: 大鼠股骨骨不連中心區(qū)在修復早期BMP2基因的低表達及表達時間滯后可能是其骨不連發(fā)生的原因之一。

Expression of BMP2 gene in different region of rat’s femoral nonunion model

[Abstract] Objective: To observe the genic dynamic expression of bone morphogenetic protein2(BMP2) during restoration of rat femoral nonunion and to discuss its influence to the restoration of rat femoral nonunion. Methods: The models of femoral nonunion of SD rats were established in day 1,3,7,14,21,28,35 before we extracted the specimens, and then the genic expression of BMP2 in different repairing stage of nonunion models were detected by RTPCT technique. Results: In different stages of femoral nonunion, the genic expression of BMP2 between central zone and outer zone also varied, their expression increment and the peak level were significantly lower in the central zone than those in the outer zone,and we can find the lag in the genic expression of BMP2 in the central zone. Conclusion: Lower value and the lag in genic expression of BMP2 in the central zone during the early stage may be an important reason of nonunion occurring in rat femur醫(yī).學全.在.線網(wǎng)站payment-defi.com.

導致骨缺損、骨不連的原因仍不完全明了。在缺損區(qū)域促進骨形成是骨不連治療的重要理念,在能夠促使骨形成的因素中,最引人注目的是細胞因子的誘導和促進作用,其中成骨作用最為突出的是骨形態(tài)發(fā)生蛋白2(bone morphogenetic protein2,BMP2)。BMP2是最主要的調(diào)節(jié)骨形成的信號蛋白,BMP2基因的表達在骨的發(fā)生、誘導、修復和骨量保持方面發(fā)揮著重要而關鍵的作用,其表達水平的高低直接影響間充質(zhì)骨細胞分化和骨量產(chǎn)生水平,而BMP2基因在骨不連中表達的相關報道較少。本實驗通過建立大鼠股骨骨不連模型探討骨不連修復過程中BMP2基因表達在時相和位相上差異,以初步了解BMP2基因在骨不連修復過程中的表達情況,探討B(tài)MP2對骨不連修復可能產(chǎn)生的影響。
