醫(yī)學(xué)免費(fèi)論文:卡瓦脂質(zhì)體納米顆粒制備及其穩(wěn)定性的研究
【摘要】 目的 制備包載卡瓦的脂質(zhì)體納米顆粒并研究其穩(wěn)定性。方法 采用旋轉(zhuǎn)薄膜蒸發(fā)法制備卡瓦納米制劑,并對其形貌、大小等進(jìn)行表征,考察納米顆粒的穩(wěn)定性。結(jié)果 卡瓦脂質(zhì)體納米顆粒平均粒徑為(99.7±7.6)nm,多分散系數(shù)為0.321±0.011,Zeta電位為(-33.9±2.3)mV,包封率為(83.32±0.34)%;納米溶液于4℃下放置60d,其納米粒外觀、粒徑及包封率無明顯變化,差異均無統(tǒng)計(jì)學(xué)意義(P>0.05)。結(jié)論 卡瓦脂質(zhì)體納米顆粒的包封率較高,粒徑分布均勻,穩(wěn)定性良好,為卡瓦的新劑型開發(fā)提供了實(shí)驗(yàn)依據(jù)。

Department of Chemistry,Guangdong Medical College,Dongguan 523808, China)Abstract:Objective To prepare the kavaloaded lipid nanoparticles from Piper methysticum and study their stability.Methods Kavaloaded lipid nanoparticles were prepared by rotaryfilm evaporation method, and their appearance, size and stability were observed.Results Mean size, polydispersity index,Zeta potential, and entrapment efficiency of kavaloaded lipid nanoparticles were 99.7 nm, 0.321,-33.9 mV, and 83.32%, respectively. Their appearance, size and entrapment efficiency showed no obvious changes after storage at 4℃ for 60 days. Conclusion The kavaloaded lipid nanoparticles have high entrapment efficiency, uniform particle size and good stability. The present study provides the experiment evidence for the development of novel dosage form of kava.

植物提取物卡瓦油是從南太平洋島嶼盛產(chǎn)的胡椒科植物卡瓦(Piper methysticum)的干燥根中提取出來的植物生理活性成分?ㄍ哂褪且环N良好的抗抑郁藥,具有鎮(zhèn)靜催眠、抗真菌、抗血栓生成、抗疲勞、減肥、松弛肌肉等作用,在全球范圍內(nèi)受到廣泛的關(guān)注[12]。但是卡瓦油的水溶性差,生物利用度低,目前尚沒有研制出理想的卡瓦藥物制劑,嚴(yán)重影響其廣泛使用。脂質(zhì)體納米顆粒是極具潛力的給藥載體,不僅能運(yùn)載親油性藥物,還能運(yùn)載親水性藥物,同時(shí)還可作為藥物的儲(chǔ)庫,增加藥物在皮膚的滯留量和滯留時(shí)間[34]。脂質(zhì)體納米顆粒制備簡單,可多途徑給藥,易于規(guī)模化生產(chǎn)。有關(guān)卡瓦脂質(zhì)體納米顆粒制備及其穩(wěn)定性研究目前尚未見有關(guān)報(bào)道,本文以卡瓦油為模型藥物來考察脂質(zhì)體納米顆粒的制備及其物理穩(wěn)定性。
