醫(yī)學(xué)論文范文:環(huán)氧化酶-2及其選擇性抑制劑塞來(lái)昔布與舌鱗狀細(xì)胞癌侵襲的研究進(jìn)展
【摘要】 環(huán)氧化酶(COX)是致炎物質(zhì)地諾前列酮合成過(guò)程中的重要限速酶。COX-2是環(huán)氧化酶的誘導(dǎo)型,參與炎癥反應(yīng)和腫瘤的發(fā)生。包括舌鱗狀細(xì)胞癌在內(nèi)的大多數(shù)惡性腫瘤均有COX-2的陽(yáng)性表達(dá)。COX-2的活性增強(qiáng)還能提高細(xì)胞與細(xì)胞外基質(zhì)蛋白的黏附能力,增強(qiáng)基質(zhì)金屬蛋白酶的表達(dá)水平,增加腫瘤細(xì)胞的侵襲力。進(jìn)一步研究COX-2選擇性抑制劑塞來(lái)昔布與舌鱗狀細(xì)胞癌侵襲之間的關(guān)系,對(duì)舌鱗狀細(xì)胞癌的治療和預(yù)后有非常重要的意義。

[Abstract] Cyclo-oxygenase(COX) is the major rate-limiting enzyme for the synthesis of prostaglandins. COX-2,which is the inducible form, involved in inflammation and cancer incidence. Study found, COX-2 was overexpressed in the majority of malignant carcinomas including tongue squamous carcinoma. In addition, the increasing activity of COX-2 can improve the adhesion capacity of cells and the extracellular matrix protein, enhance expression level of matrix metalloprotease, and increase the invasion of tumor cells. 舌鱗狀細(xì)胞癌(tongue squamous cell cacrci-noma,TSCC)是口腔頜面部最常見(jiàn)的惡性腫瘤之一,早期即可發(fā)生頸部淋巴結(jié)轉(zhuǎn)移。舌鱗狀細(xì)胞癌轉(zhuǎn)移嚴(yán)重影響患者的預(yù)后,也是舌鱗狀細(xì)胞癌患者死亡的重要原因之一。大量的研究資料顯示,環(huán)氧化酶(cyclo-oxygenase,COX)-2通過(guò)多種機(jī)制參與惡性腫瘤的發(fā)生發(fā)展和浸潤(rùn)轉(zhuǎn)移,而其選擇性COX-2抑制劑塞來(lái)昔布對(duì)多種惡性腫瘤的發(fā)生和發(fā)展都有明顯的抑制作用。COX-2在舌鱗狀細(xì)胞癌中呈過(guò)度表達(dá),其水平明顯高于癌前期病損黏膜和正常上皮。下面就該酶在舌鱗狀細(xì)胞癌侵襲中的作用及其特異性抑制劑塞來(lái)昔布對(duì)舌鱗狀細(xì)胞癌侵襲作用的抑制作一綜述。
